MinION DNA barcoding of five representative species

To test the accuracy and the efficiency of DNA barcoding with MinION nanopore we amplified the relevant barcode regions from the genomic DNA of five representative species Escherichia coli (16S rRNA gene), Homo sapiens (COI), Gracilaria cearensis (18S rRNA gene), Ulva fasciata (tufA) and Saccharomyces cerevisiae (ITS), representing bacteria, metazoans, green and red algae as well as fungi, respectively. The amplicons were sequenced with MinION using ligation protocol with multiplexing that was followed by a bioinformatics pipeline that was adapted from Maestri et al. (2019)
