Genetic and pharmacological evidence for kinetic competition between alternative poly(A) sites in yeast - NNS complex

PAT-seq approach was used to determine changes to 3'UTR length in yeast upon mutation of NNS subunits Nrd1 (nrd1-5), Nab3 (nab3-11) and Sen1 (sen1-1) compared to wild type cells Overall design: Yeast cells were grown at 25ºC until an OD600 of 0.6 was reached then transferred to 37ºC for 1 hour before harvesting. Samples were analysed via PAT-seq 3'UTR focused deep-sequencing in replicate using the Illumina Hiseq1500 platform.
