The proposed experiment is addressed to use the MARI spectrometer to study the thermal denaturation process of the psychrophile enzyme alpha-amylase in presence of bioprotectant mixtures composed by trehalose and glycerol mixtures. In particular it is expected that the action of trehalose/glycerol mixtures would lead to a shift of the unfolding process towards higher temperatures, its spreading over a larger temperature range, together with a strengthening of the solute-solvent hydrogen bonded network. By performing runs of six hours on alpha-amylase in trehalose/glycerol mixtures at four concentration values in the range 0-10% and for six temperature values in the temperature range 310 K and 390 K and the necessary calibration and empty cell measurements, we estimate a total of 7 days of beam time on MARI for the required statistics.
