Heterogeneity in the transcriptional response of the human pathogen Aspergillus fumigatus to the antifungal agent caspofungin

The transcription factor CrzA influences cell wall organization in the pathogenic fungus Aspergillus fumigatus, and also binds to the promoter regions of chitin synthase genes upon exposure to the antifungal drug caspofungin. To gain an overview of the genes directly regulated by CrzA, the CrzA binding sites were determined genome-wide by ChIP-seq Overall design: The CrzA-tagged strain was grown for 16 hours at 37oC in minimum liquid media at 220 rpm and then added with 2 ug/ml of caspofungin for 1 hour minutes (37oC 220 rpm). After crosslinking, the cells were washed and frozen for chromatin extraction and immunoprecipitation.
