This dataset mainly contains experimental and fitting data to quantify the dynamics of lipids of mimetic membranes in presence of a glycan layer at the nanoscale.
The experimental data is composed of AFM images (.ibw), with selected force maps (.ARDF) , confocal FCS images (.tif) and FCS traces (.dat).
The raw data were analyzed in the Symphotime program of the FCS setup (.dat). Further anylsis was done in Matlab (.mat) which required input data saved in .xlsx format. The images shown in the publication were all exported into the .png format.
From the AFM images and the confocal FCS images height profiles have been obtained. From the force maps the typical forces were pooled into a bimodal distribution of forces. The FCS traces were fitted to obtain characteristic diffusion times.
Data processing methods:
The processing was performed using Matlab and the software of the AFM microscope and the FCS setup (Symphotime).
Information about instruments, calibration and standards:
stated in the paper
Environmental or experimental conditions:
All measurements were performed at normal temperature and pressure. The data was analyzed on standard 16 GB office PCs.
Quality-assurance procedures performed on the data:
The confocal FCS setup has to be calibrated to yield the maximum possible photon counts per second and the AFM setup requires a calibration of the tip in use at each experimental day. At the beginning of an experimental day, it was checked that the lipid layer is a single layer by a signle force measurement.
