Replication Data for: Improved Biostimulant Production through Solid-State Fermentation of Green Waste Using Low-Temperature Strategy in Tray Bioreactors

Raw data from a series of experiments aimed to develop solid-state fermentation (SSF) process to increase the production of biostimulants (indole-3-acetic acid-IAA) in a tray bioreactor using a temperature strategy. The process was conducted by Trichoderma harzianum, utilizing green waste as substrate.

METHODOLOGICAL INFORMATION

The experiments was involved Solid state fermentation (SSF) using Trichoderma harzianum as inoculum and green waste (grass clipping) as substrate. The production of IAA and conidial spores was studied in a tray reactor. A PCA and GLMM analysis were performed. The effect of a low temperature strategy was tested to increase the IAA production.

  1. Description of methods used for collection-generation of data:

A solid state fermentation was conducted in a tray bioreactor. The mixture of green waste and bulking agent (wood chips) was inoculated by Trichoderma harzianum. The fermentation was 10 days. In each experiment one single trya was used and tThe total wet weight of material loaded in tray was 500 g. The material aerated with the airflow of 2.9 - 3.7 mL min⁻¹ g⁻¹ DM. 10 g of solid samples taken from the tray reactor in specific days during fermentaion was mixed with 50 mL Tween 80 solution (0.1%) in Erlenemeye fasks. After 30 min agitation, the liquid phase used for spore and IAA analysis.

-Bio pesticide measured as conidial spores of Trichoderma which counted using a Neubauer Chamber and microscope. -IAA was quantified by HPLC-UV. -Oxygen consumption was measured as specific oxygen uptake rate (sOUR) using a system developed by the group

  14. Tichpderma harzianum CECT 2929 was cryopreserved at -80°C.

  15. Microorganism cultured in malt extrat agar at room temperature for 6-8 days.
  16. Fermentations was conducted in tray reactor at room temperature.

  17. Quality-assurance procedures performed on the data:

During each fermentation, samples were on days taken 3, 5, 7, 10, 13, and 15. At each time point, two independent samples were taken from randomly selected spots on the tray. The analysis of IAA and spores was carried out in triplicate from each samples.
