Antibody BAM1 binding signals and deoxyhexose concentrations in anion exchange fractions of glycans extracted from sediment cores PS097/72-2 and PS097/72-1

Sediment sections from cores PS097/72-2 and PS97/072-1 collected on the expedition PS97 were freeze-dried and homogenized. Glycans were sequentially extracted from the freeze-dried sediment as described by Hellige et al., 2026. Simultaneously, replicates of combusted sand were treated as the negative controls. The extracts were pooled and loaded onto an anion exchange column to isolate anionic glycans. By applying an increasing salt gradient, the extracted glycans were fractionated into 30 x 1 mL AEX fractions. The anion exchange fractions were probed for the presence of fucoidan with monoclonal antibody (BAM1; Torode et al., 2015) using enzyme-linked immunosorbent assay (Hellige et al., 2026). Additionally the deoxyhexose concentrations of the fractions were also quantified using a modified L-cysteine assay (manuscript in prep, Dische and Shettles, 1948). The deoxyhexose concentrations were normalized to the weight of dried sediment.
