CYCLODEXTRIN-ENCAPSULATION OF PLATINUM-BASED ANTICANCER DRUGS

The goal of the present proposal is the study of supramolecular nanoencapsulation of platinum-based anticancer agents, by INS spectroscopy. A chemically modified cyclodextrin (hydroxylpropyl-beta-cyclodextrin) will be used as a carrier for cisplatin and cisplatin-like mono- and dinuclear polyamine chelates. Both inclusion complexes and physical mixtures will be investigated, in order to monitor the spectral changes induced by host-guest interactions. In addition, the cyclodextrin-protecting ability towards drug-glutathione binding (responsible for an unwanted acquired drug resistance) will be assessed, by analysis of inclusion complexes formerly incubated with a glutathione excess. The results thus gathered will be interpreted in the light of theoretical and optical vibrational data obtained for these systems, as well as INS spectra previously acquired by the team for the Pt-drugs.
