Amplicon sequencing of 18S rRNA gene, eukaryote assemblages in seawater in Matsushima Bay, Japan

To understand whether a tsunami in Matsushima Bay, Japan caused environmental changes, we analysed seawater from 2004 to 2005 and from 2012 to 2016. After DNA was extracted, the 18SrRNA gene were amplified with primers (TAReuk454FWD1 and TAReukREV3) using PCR. The PCR product mixtures were analyzed for pair-end DNA sequencing of 300 bp Ã? 2 using MiSeq desktop sequencer (Illumina). The sequenced data were analyzed in the Qiime bioinformatics pipeline.
