Sanger sequencing raw data and mutation analysis of mutants of lambda's cII coding sequence, obtained by fluctuation assay and mutation generation. Contains a zipped file with all traces files (.ab1) that were used in the analysis, a fasta file conntaining the reference sequence used to analyse the trace, a tab-separated file containing the correspondance between trace files and the experiment associated and a tab-separated file containing the fluctuation assay informations required for mutation rate estimation, using the Ma, Sandri and Sarkar maximum likelihood, P0 and generating function estimators (see Foster, Methods Enzymol. 2006 ; 409: 195–213). The trace file were analysed using Clone Manager v10.1 and the reference sequence NC_001416.1:38360-38653 (NCBI), by using the subclonal sequence assembly tool.
The traces files are kept as raw data due to a mixture of mutants and WT phenotypes in the sequenced samples, resulting in double pics for some mutants. These mutations would be regarded as low sequencing quality by a connsensus making tool for conversion into a fastq file.
