Sanger sequencing of lambda cII mutants, obtained by fluctuation assay

Sanger sequencing raw data and mutation analysis of mutants of lambda's cII coding sequence, obtained by fluctuation assay and mutation generation. Contains a zipped file with all traces files (.ab1) that were used in the analysis, a fasta file conntaining the reference sequence used to analyse the trace, a tab-separated file containing the correspondance between trace files and the experiment associated and a tab-separated file containing the fluctuation assay informations required for mutation rate estimation, using the Ma, Sandri and Sarkar maximum likelihood, P0 and generating function estimators (see Foster, Methods Enzymol. 2006 ; 409: 195–213). The trace file were analysed using Clone Manager v10.1 and the reference sequence NC_001416.1:38360-38653 (NCBI), by using the subclonal sequence assembly tool. The traces files are kept as raw data due to a mixture of mutants and WT phenotypes in the sequenced samples, resulting in double pics for some mutants. These mutations would be regarded as low sequencing quality by a connsensus making tool for conversion into a fastq file.
