This dataset characterizes various N-Acetylglucosamine oxidase (NagOX) constructs fused with different affinity and solubility-enhancing tags (6xHis, SUMO, CBM3, and CBM9) to evaluate their hydrogen peroxide (H2O2) production. 6xHis-SUMO-NagOX and 6xHis-SUMO-CBM3-NagOX were selected for comparative immobilization and purification studies, using the absence of catalase activity as a key selection criterion. 6xHis-SUMO-CBM9-NagOX was excluded from further testing due to observed proteolysis, which compromised the enzyme's structural integrity. Ultimately, the CBM3/Cellulose system (Perloza MT100 medium) yielded a higher purification factor and superior immobilization results, as it successfully eliminated catalase from the final biocatalyst.
