Single_Nematode_R_D

This study forms part of the Institute's new Tree of Life (ToL) programme. The primary goal of ToL is to sequence novel genomes representing all eukaryotic biodiversity in the UK. This project will provide reference genomes for the global scientific community that will inform a broad range of major issues facing humanity, such as the impact of climate change on biodiversity, and the conservation of endangered species and ecosystems. A major focus of the Blaxter Faculty is to provide high quality reference genome sequences and formal species descriptions of >260 species of Nematode from 53 genera. Nematodes that have been cultured in-house will be implemented in low-input trials to generate long read, linked-read, complementary DNA and HiC libraries for individual nematodes. The Nuclei EXtraction by SONication (NEXSON) method will be trialled to disrupt the nematode cuticle, isolating nuclei that will be split and prepared for HiFi, linked-read and HiC sequencing. The development of this method has particular implications for our ability to generate chromosomal assemblies for free-living and parasitic nematode species which cannot be easily cultured.
