Early stages of amyloid aggregate formation

We will study the kinetics of the early stages of aggregate formation in protein solutions under conditions that are known to result in the formation of amyloid protein fibres and spherulites. These structures are often linked to the disease pathologies of conditions such as Alzheimer's and Type II diabetes. Recent studies indicate that the small species that form prior to these higher aggregates are actually the main cytoxic species. Light scattering studies indicate that objects with sizes in the range 3-100nm form in solution during the early stages of aggregation. Little is known about these initial species and about what determines the final balance of fibrils and spherulites in solution. We will vary the protein concentration in a range where fibrils and spherulites are known to form and will collect time resolved SANS data to obtain info about the size/shape of these small objects
