Interaction of Low Density Lipoproteins (LDL) with Model Mammalian Cell Membranes

Atherosclerosis and subsequent clinical complication is one of the biggest killers in the western world, with a death toll reaching 16.7 million deaths per year. Although many risk factors have been identified, it is well known that various lipoprotein particles in the blood play an important role in the development and rupture of atherosclerotic plaques. Upon adsorption to the cell membrane LDL is thought to exchange both lipids and cholesterol with the blood vessel wall. The unloading of LDL cholesterol cargo may initiate atherosclerotic plaque development. In this experiment we aim to study firstly the adsorption of LDL to a supported lipid bilayer as a function of bilayer charge (PC:PS) ratios. Secondly, through the use of fully deuterated lipid molecules (DMPC, DMPS and cholesterol) we aim to study exchange of the LDL components with the bilayer.
