RNA Sequencing analysis of Arabidopsis thaliana seedlings and flower buds after chemical inhibition of TOR Kinase

To elucidate localised downstream targets of TOR Kinase in different tissues, 100uM of the TOR inhibitor, Torin2, was exogenously applied to Arabidopsis thaliana seedlings 5 days after germination, and to flower buds emerging from the central rosette. As a control, mock solvent solution was applied. Tissue was harvested 6 hours later and RNA extracted. 1ug RNA was sent for Illumina HiSeq 4000 Next Generation Sequencing. The data was analysed with FastQC, Kallisto, and DESeq2. Mock and Torin2 treated tissues were compared in the seedlings and in the flower buds. Whilst the seedling data suggest TOR Kinase activity was inhibited, resulting in changes in expression of growth and development genes, it is unclear whether the chemical treatment of flower buds was sufficient to inhibit TOR.
