Investigation on the structural properties of doxorubicin-loaded PEGylated liposomes (Doxil®)

Doxil® is a marketed liposomal formulation containing the anticancer drug doxorubicin. Recently, SAXS measurements have evidenced a crystalline fibrillar structure for the encapsulated doxorubicin at 25°C. We have already performed a characterization of the thermal behaviour of the formulation by microcalorimetry, and high-resolution ultrasonic spectroscopy in order to investigate better the phase transition of the liposomal membrane and of the encapsulated drug. We propose here to perform SANS measurements on Doxil® and unloaded liposomes for comparison at four selected temperatures. As previously verified by preliminary spectra at 20°C, this technique, thanks to H2O/D2O sub-phase matching, is able to discriminate between the contribution of the phospholipidic components and drug nanocrystals. Thus, it represents a valid tool for the characterization of the thermal behaviour of Doxil®.
