Dissolved organic carbon (DOC) and total dissolved nitrogen (TDN) concentrations and the fluorescence and absorbance properties of incubated lake and pond water, Cape Bounty Arctic Watershed Observatory (CBAWO)

East lake water was collected and filtered using a glass filtration apparatus and precombusted 0.7 μm GF/F glass fibre filters. Sample water was then distributed into 40 mL amber and clear EPA vials. Sediment from an on-site active layer detachment scar was added to the vials in three concentrations; 100, 300 and 500 mg/L. Control vials had no added sediment. Vials were incubated using wire comparments set up with attached temperature and light loggers in each pond. After 0, 2, 7, 14 and 27 days from the start of the incubation, vials were removed from each pond and filtered on site. In-situ pond samples were also taken on these days using amber HDPE bottles and were subsequently filtered as well. The samples were stored cool and in the dark until later analysis at Queen's University. DOC concentrations were measured as non-purgeable organic carbon via high temperature combustion (720°C) with a Shimadzu TOC-L analyser using a high sensitivity platinum catalyst in line with a total nitrogen module (TNM-L unit). TDN concentrations were measured as nitrogen monoxide via thermal decomposition. UV-visible absorbance and fluorescence measurements and emission excitation matrices (EEMs) were collected at room temperature using a Horiba Aqualog.
